Resumen
Antecedentes. La nitrofurantoína (NIT) es un antimicrobiano de primera línea para infecciones urinarias no complicada; la variabilidad interindividual en eficacia y seguridad podría tener un componente genético.
Objetivo. Describir y contextualizar las variantes asociadas a “respuesta a nitrofurantoína” registradas en el GWAS Catalog (rasgo OBA_2040118).
Métodos. Estudio secundario de registros del GWAS Catalog (descarga: 24-oct-2025), filtrando el rasgo “trait in response to nitrofurantoin”. Se extrajeron rsID, región génica mapeada, odds ratio (OR), intervalo de confianza (IC95%), valor p y tamaño muestral reportado por el estudio fuente. Se ordenaron por evidencia estadística y se generó un resumen descriptivo.
Resultados. Se identificaron 4 asociaciones provenientes de un solo estudio (PMID 28043905; Gastroenterology, 2016) con 64 casos europeos y 10,588 controles europeos. Las señales correspondieron a: rs72696020 (HISLA, OR 7.8; p=6×10⁻⁸), rs6694270 (PAX7–TAS1R2, OR 2.59; p=2×10⁻⁷), rs10404821 (C19orf81, OR 2.56; p=2×10⁻⁷) y rs61858823 (NEK4P3–MYL6P3, OR 5.63; p=5×10⁻⁷). La frecuencia del alelo de riesgo no estuvo disponible (NR).
Limitaciones. Única cohorte de descubrimiento, ascendencia europea exclusiva y valores p que se aproximan o superan marginalmente el umbral clásico de GWAS, con ausencia de replicación.
Conclusión. Las asociaciones actuales sugieren loci candidatos para susceptibilidad a eventos adversos/efecto de NIT, pero requieren replicación y caracterización funcional antes de cualquier aplicación clínica o farmacogenómica.
